 Introduction

The gene-trapping principle is based on the random integration of a reporter/selector cassette into a genome in order to simultaneously select for, mutate and identify the trapped locus.

Splice Acceptor Gene - Trapping

Splice Acceptor Gene - Trapping
A: wildtype locus

B: trap-vector integration

Typical gene-trap vectors are promoterless and function by generating a fusion transcript with the endogenous gene. The presence of splice donor or acceptor elements leads to the generation of a fusion protein, even after intron integrations.
